Gene targeting strategy for B-hIL10 mice. The exons 1-5 of mouse Il10 gene that encode the full-length protein were replaced by human IL10 exons 1-5 in B-hIL10 mice.
Strain specific analysis of IL10 gene expression in WT and B-hIL10 mice by RT-PCR. Mouse Il10 mRNA was detectable in splenocytes of wild-type mice (+/+), Human IL10 mRNA was detectable only in homozygous B-hIL10 mice (H/H) but not in WT mice.
Serum from both wild type (WT) C57BL/6 and homozygous B-hIL10 mice were analyzed by ELISA. When treated with LPS, mouse IL10 was only detectable in WT C57BL/6 mice, while human IL10 was only detectable in homozygous B-hIL10 mice. ND, not detectable. n=3.
